The current study investigates the effect of Aqueous Extract of Moringa oleifera (AEMO) Â leaf on the oxidative stress, antioxidant status and histopathological changes in testes of rats underÂ insulin deficient condition. Male Wistar rats were used in this study. Diabetes (insulin deficient)Â was induced in rats by a single intraperitoneal injection of Streptozotocin. AEMO was administeredÂ atÂ aÂ dose ofÂ 200Â mg/kgÂ bodyÂ weightÂ forÂ a periodÂ ofÂ 60Â days. At theÂ endÂ ofÂ theÂ experimental period,Â theÂ rats wereÂ sacrificedÂ by cervicalÂ dislocation.Â RelativeÂ organÂ weightÂ wasÂ calculated.Â OxidativeÂ stressÂ wasÂ assessedÂ byÂ theÂ extentÂ ofÂ lipidÂ peroxidationÂ (LPO)Â andÂ theÂ levelsÂ ofÂ reducedÂ GlutathioneÂ (GSH) inÂ testes.Â The statusÂ of antioxidantÂ enzymesÂ was estimatedÂ by determingÂ theÂ activitiesÂ ofÂ GlutathioneÂ ReductaseÂ (GR),Â Glutathione-Â S-TransferaseÂ (GST),Â SorbitolÂ dehydrogenase (SDH),[CatalaseÂ (CAT)Â andÂ SuperoxideÂ dismutaseÂ (SOD).Â HistologicalÂ changes inÂ testesÂ wereÂ studied.Â StatisticalÂ significanceÂ ofÂ theÂ resultsÂ was evaluated byÂ Duncanâ€™s MultipleÂ RangeÂ Test.Â InÂ thisÂ study,Â diabeticÂ ratsÂ showedÂ significantÂ decreaseÂ in
absoluteÂ and relativeÂ weightsÂ ofÂ testes,Â increasedÂ LPOÂ andÂ decreasedÂ levels
TheÂ activitiesÂ ofÂ antioxidantmÂ enzymes-Â GR,Â GST,Â SOD, CATÂ wereÂ reduced. InÂ contrast,Â theÂ activity ofÂ SDH,Â a polyolpathwayÂ enzyme,Â increased.Â HistologicalÂ pictureÂ ofÂ testesÂ of DÂ groupÂ showedÂ pathologicalÂ changes.Â However,Â administrationÂ of AEMOÂ showed beneficialÂ effectÂ whichÂ wasÂ reflectedÂ by reducedÂ oxidativeÂ stress,Â enhancedÂ activitiesÂ ofÂ antioxidantÂ enzymesÂ inÂ testes.Â AEMOÂ alsoÂ protectedÂ theÂ testesÂ againstÂ theÂ histological changesÂ inducedÂ byÂ oxidativeÂ stressÂ underÂ insulinÂ deficientÂ condition.Â TheÂ studyÂ revealedÂ that AEMOÂ amelioratedÂ oxidativeÂ stress,Â exhibitedÂ antioxidantÂ potential,Â andÂ playedÂ a roleÂ inÂ theÂ protectionÂ ofÂ testicularÂ structuralÂ integrityÂ underÂ insulinÂ deficientÂ condition.
